アーティファクト An artifact

先日、勤務先の別グループの先生から新井先生にマウス切片を診てほしいとのご依頼がありました。遺伝子改変マウス脳のPFA固定、パラフィン包埋切片です。私も同席し標本を観察しました。すると、切片全体に空胞が散在しているのが目を引きます。新井先生曰く、空胞は血管ではないかとのこと。空胞は特異的な部位にあるわけではなく切片全体にありますし、また、正常マウス脳にもあるので病変ではなさそうです。自分で作製した標本ではないので断言は出来ませんが、固定もしくはパラフィン包埋過程で発生したアーティファクトと考えられます。さらにご依頼があれば、技術的にご支援したいと考えています。

The other day, Dr. Arai received a request from researchers in another group at my workplace to examine a mouse section. It is a PFA-fixed and a paraffin-embedded section of a genetically modified mouse brain. I also attended and observed the specimen. Then, it is noticeable that vacuoles are scattered throughout the section. According to Dr. Arai, the vacuoles may be blood vessels. The vacuoles are not in specific sites but in the entire section. They are also found in normal mouse brains, so they are unlikely to be lesions. I can't say for sure because it is not a specimen that I prepared, but it should be an artifact generated during the fixation or paraffin embedding process. If they have any further requests, I am willing to provide technical support.
